Abstract

Objective To investigate the effects of sodium butyrate on cell growth inhibition and apoptosis in PC-3M cell lines. Methods Sodium butyrate was added to the culture media of PC-3M cells for different hours and then the cell changes were observed by phase-contrast microscope, the inhibitory rate of cell growth was detected by MTT assay, the apoptosis rate was determined by AO/EB assay, the apoptosis peak and the changes of cell cycle were measured by flow cytometry (FCM). Results Cell growth was significantly inhibited by sodium butyrate and the inhibitory effect was in time- and dose-dependent manner. The morphological changes of apoptosis in PC-3M cells were found. The cell cycle was arrested to G 1/G 0 and G 2/M, the number of cells in S phase reduced, sub-G 1 appeared. Conclusion Sodium butyrate can inhibit the cell growth and induce apoptosis in PC-3M.
