Abstract

Objective To determine the content of total flavonoids and puerarin in Radix Puerariae thomsonii of different originations.Methods The content of total flavonoids was determined by ultraviolet spectrophotometry(UV).The content of puerarin was determined by HPLC,the analysis was performed on a kromasil C18 column(4.6 mm×250 mm,5μm) with methanol-water(33:67) as mobile phase at a flow rate of 1.0 ml·min-1,and at a column temperature of 30℃.The detection of wavelength was 250 nm.Results The content of total flavonoids was 0.53~1.03%,and recovery rate was 99.69 %,RSD2%(n = 6).The content of puerarin was 0.21%~049%,and recovery rate was 99.11 %,RSD2%(n = 6).Conclusion The content of total flavonoids and puerarin in Radix Puerariae thomsonii has great changes between the different originations,the contents of total flavonoids and puerarin in Guangdong Radix Puerariae thomsonii are higher,and the average quality is better than Guangxi Radix Puerariae thomsonii.
