Effects of ligustrazine on NO and iNOS in rats with acute hepatic injury induced by Paraquat

Abstract

Objective To investigate the effects of ligustrazine on nitric oxide(NO) and inducible nitric oxide synthase(iNOS) in rats with acute hepatic injury induced by Paraquat.Methods 50 Sprague-Dawley(SD) rats were randomly divided into five experimental groups: blank group,negative control group,positive control group,ligustrazine low-dose group(LDG),ligustrazine high-dose group(HDG).Macroscopic and histopathological examinations were performed.In the mean time,the level of ALT,AST,MDA,SOD and GSH-Px in plasma were detected,and the activities of hepar tissue homogenate NO and iNOS were measured.Results Compared with that in negative control group,ALT,AST,MDA,SOD and GSH-Px in positive control and ligustrazine low-dose group had statistical significance(P0.05,0.01),and biological indexes of hepatic injury and the contents of NO and iNOS were significantly decreased(P0.05,0.01).Hemorrhage of liver of ligustrazine low-dose group were mitigated.But that in ligustrazine high-dose group don't have statistically significant.Conclusions Ligustrazine has protective effect on hepatic injury induced PQ and their therapeutic mechanism is possibly via inhibiting the activities of NO and iNOS.
