Comparative analysis of platelet count between hematology analyzer XE-2100 and blood smear reviews

Abstract
Objective To analyze the relationship of count accuracy between hematology analyzer XE-2100 and blood smear reviews. Methods The precision,linearity and correlation of count were determined by hematology analyzer XE-2100,CD-3700 and manual method respectively; The samples with count flags out of CD-3700 were selected to be measured again by both XE-2100 and manual method; the agreement rates of count by hematology analyzers with the different concentration were compared separately those under microscopy. The morphology of blood cells and white blood cells on blood smears were also referred when the flags showed on the analyzers. Results On hematology analyzer XE-2100, the precision of count showed high with the coefficient variation of 2% and fair linearity. The linear correlation of count was γ=0.9 between the methods by PLT-O or PLT-I on XE-2100 and method by microscope. The more the concentration reduced,the more the agreement of count decreased between XE-2100 and blood smears. The flags platelet clumps or distribution indicated on XE-2100 were related to the increased large platelet, red blood fragment and small blood cell on blood smears. The agreement of count flags between the methods by PLT-O and PLT-I on XE-2100 reached the highest when the concentrations were normal range. Conclusion The hematology analyzer XE-2100 showed high accuracy count. Microscope reviewing is necessary when the abnormal scatergrams and flags displayed on XE-2100 or abnormal concentration.
