Abstract
In this paper,we used mature dry seeds of four maize elite inbred lines(liao7980,dan9818,340,5026) as explants,some important factors affected induction,maintenance and differentiation of callus from mature embryos were investigated.The results indicated that genotype has remarkable effect on embryogenic callus induction,2,4-Dichlorophenoxyacetic a cid(2,4-D) was a critical factor for producing primary callus.Supplement with optimal L-pro in the subculture medium significantly promoted the formation of embryogenic callus but growth rate was not significantly different from the control.And the addition optimal level of BA and AgNO3 into regeneration medium has increased the frequency o f plant regeneration.
