Effect of C-peptide on proliferation of rat mesangial cells cultured in high glucose

Abstract

Objective To evaluate the effect of different concentrations of C-peptide on the proliferation of rat mesangial cells line HBZY-1 cultured in vitro in high glucose.Methods Rat mesangial cells line HBZY-1 were treated in different groups. Control group (1ow glucose l g/L without C-peptide) and experimental groups (high glucose 6 g/L with C-peptide 0,0.5,10, 50, 300 and 900 nmol/L) were established and incubated for 24, 48 and 72 hours. The proliferation effect of C-peptide on cells was measured with MTT method. Results High glucose concentration significantly enhanced the proliferation of HBZY-1 rat mesangial cell.The different concentration of C-peptide on high glucose concentration induced mesangial cell proliferation to varying degrees.Cells treated for 24 hour with 10,50,300 nmol/L of C-peptide significantly inhibited the proliferation(P0.05),while 0.5,900 nmol/L showed no significant role. Conclusion C-peptide has an intrinsic capability of inhibiting the proliferation of rat mesangial cells line HBZY-1 in cultured high glucose and these effects are in a C-peptide concentration and time dependent manners. The reasonable application of C-peptide may be helpful in preventing and treating diabetic nephropathy.
