Abstract

Objective To investigate the effects and mechanism of sodium ferulate(SF)on the proliferation of mesangial cells induced by high glucose.Methods The primary glomerular mesangial cells were divided into normal,mannitol,high gloucose and high gloucose plus different concentration SF groups.Cells were collected at 24 and 48 h.Cell proliferation was measured by MTT assay,and cell cycle and cyclinD1 were measured through flow-cytometry.p53 protein expression was detected by immunocytochemistry.Results High glucose significantly promoted mesangial cell proliferation and up-regulated the expression of cyclinD1,and down-regulated expression of p53 within 48 h.SF inhibited mesangial cell proliferation induced by high glucose with concentration and time-depended manner.Conclusions SF can inhibit proliferation of mesangial cells induced by high glucose by suppressing the cyclinD1 expression and increasing the level of p53 protein possibly.
