Abstract
[Objective] The purpose was to study the optimum culture condition for culturing Tremella aurantialba mycelia by liquid-submerged fermentation and the extraction technology of its polysaccharide.[Method] The optimum culture condition for culturing T.aurantialba mycelia by liquid-submerged fermentation was ascertained through screening media and optimizing culture condition and the extraction effects of alkali extraction method and hot water soaking method on T.aurantialba polysaccharide were studied through comparison and analysis.[Result] When the corn powder,bran and sweet potato powder were used as carbon sources,the yields of T.aurantialba mycelia were 19.1,13.2 and 10.9 g/L resp.When the addition amounts of corn powder were 1%,2%,3%,4% and 5%,the yields of T.aurantialba mycelia were 8.6,17.1,19.5,15.4 and 7.2 g/L resp.When the peptone,yeast paste,urea and ammonium sulfate were used as nitrogen sources,the yields of T.aurantialba mycelia were 18.9,10.3,6.7 and 5.2 g/L resp.When the addition amounts of peptone were 0.2%,0.4%,0.6%,0.8% and 1.0%,the yields of T.aurantialba mycelia were 10.1,15.7,23.0,20.3 and 18.7 g/L resp.The yields of crude polysaccharides and polysaccharide extracted by alkali extraction method were increased by 6.9% and 36.4% resp.in comparison with hot water soaking method.[Conclusion] This experiment supplied technical support for the production of T.aurantialba mycelia and the extraction of its polysaccharide.
