Abstract

Objective:An RP-HPLC method was applied to determine puerarin,daidzin and daidzein in Radix Pu- erariae Thomsonii of different cultivated species,and to develop a good method to control the quality for Radix Puer- ariae Thomsonii.Methods:An RP-HPLC method was applied with a Diamonsil C_(18)column(150 mm×4.6 mm, 5μm)by gradient elution using methanol-1% acetic acid solution as the mobil phase(0-13min,25:75;13-15 min,40:60;15-30min,60:40;30-40 min,25:75).The flow rate was 1 mL·min~(-1).The detective wavelength was 250nm.The column temperature was 25℃.The sample injection volume was 5 μL.Results:Three compounds showed good linearity(r0.9995)in a detected concentration range.The average recovery rate was in the range of 99.6%-100.9%.The RSD of precision was less than 2%.The contents of puerarin,daidzin and daidzein in Xi- aoye Fenge were the highest in the three determined species.Conclusion:The method is simple,accurate and can be used to control the quality for Radix Puerariae Thomsonii.
