Abstract
Objective To observe the effect of growth inhibition and apoptosis on human Burkitt lymphoma carcinoma cell line Raji induced by oxaliplatin.Methods Raji cells were incubated with oxaliplatin at various concentration(0、6.25、12.5、25、50、100 μg·mL-1)and various length of time(24,36,48 hours),cell growth inhibition was assessed by CCK-8 methods,cell-cycle kinetics and apoptosis was analyzed by fluorescence microscopy and flow cytometry.Results The growth of Raji cells was significantly inhibited by oxaliplatin in a dose-dependent and time-dependent fashion.The inhibition rate of cell growth was(94.73±1.40)% after incubation with 100 μg·mL-1 oxaliplatin for 48 hours.Oxaliplatin could cause Raji cells cycle to change.With the increase of oxaliplatin concentration,G2/M phase cell percentage increased.The early-stage and late-stage apoptosis rate of Raji cells were(8.25±1.79) % and(14.61±2.18)% respectively after incubation with 100 μg·mL-1 oxaliplatin for 48 hours.Conclusions Oxaliplatin suppresses the growth of Raji cells in vitro by causing cell-cycle arrest and cell apoptosis.
