Abstract

Objective To investigate the application of proteomics in the field of serology, and to screen the differential expression proteins related with gastric carcinoma. Methods Two-dimensional electrophoresis was applied to segregate the total proteins in the serum form gastric cancer patients and health volunteers. After staining, the differential expression proteins were analyzed using ImageMaster 2D Elite software, and identified using matrix-assisted laser desorption/ionization-time of flight mass spectrometry(MALDI-TOF-MS).Results Electrophoresis figures with high reproducibility were obtained. The four high-abundance gastric cancer protein spots were identified and analyzed. The special protein spots were Serpin B6(Placental thrombin inhibitor)(Cytoplasmic antiproteinase)(CAP)(Protease inhibitor6)(PI-6),Septin-1(LARP)(Serologically defined breast cancer antigen NY-BR-24),Kallikrein-6 precursor(Protease M)(Neurosin)(Zyme)(SP59),Hemoglobin beta chain. Conclusions Protein expression is differenti in the serum from the gastric cancer patients and health volunteers. It is hopeful to find the biomarkers related with poorly differentiated gastric carcinoma using proteomics.
