Abstract

Objective To establish two-dimensional electrophoresis(2-DE)profiles of human gastric cancer tissue and paired normal tissue,and to identify differential expression proteins,in which try to find out candidates as gastric cancer-related proteins.Methods 2-DE was applied to separate the total proteins of gastric cancer tissue and paired normal tissue.After staining and analyzing by PDQuest soft- ware,the differential expression proteins were identified by matrix-assisted laser desorption/ionization- time of flight mass spectrometry(MALDI-TOF-MS)or MALDI-TOF-TOF-MS.Results 2-DE profiles with high resolution and reproducibility were obtained.Twenty-three protein spots were found as differ- entially-expressed proteins,in which 15 protein spots were identified successfully.Among the identified proteins,there were 10 over-expressed and 5 under-expressed proteins in gastric cancer tissue as compared with normal tissue.Conclusion The well-resolved,reproducible 2-DE patterns of human gastric cancer tissue and paired normal tissue were established and fifteen differentially expressed proteins were identi- fied.
