Effect of TSG on the Expression of MCP-1 in Vascular Endothelial Cell Induced by H_2O_2

Abstract

Objective To study the effect of 2,3,5,4′-tetrahydroxystilbene-2-O-β-D-glucoside(TSG) on expression of monocyte chemoattractant protein-1(MCP-1) induced by H2O2 on human umbilical vein endothelial cells(HUVEC).Methods Endothelial cells were cultured in vitro and divided into group of control,H2O2(200 μmol/L),simvastatin(5 μmol/L simvastatin plus 200 μmol/L H2O2) and TSG(1 μmol/L TSG plus 200 μmol/L H2O2),after treated for 24 h,the expression of MCP-1 mRNA and protein were detected by reverse transcriptase polymerase chain reaction(RT-PCR) and enzyme-linked immunosorbent assay(ELISA) respectively.Results Treating endothelial cells with 200 μmol/L H2O2,the expression of MCP-1 mRNA and protein level was significantly higher(P0.01).TSG can inhibit the expression of MCP-1 mRNA and protein level induced by H2O2 on endothelial cells(P0.05).Conclusions TSG can inhibit the expression of MCP-1 induced by H2O2 in human umbilical vein endothelial cells.
