Abstract

Objective To observe the inducing effect of cytokines on apoptosis of tumor cells when interfe ron a(IFN a) and tumor necrosis factor a(TNF a) were applied alone or together with chemotherapeutical drugs.Methods The inducing effects of cytokines on apoptosis of tumor cells(HL60,L 1210 and H22) in vitro were observed by means of optical microscope,fluoroscence microscope,electron microscope and flow cytometry. Results The typical characteristics of apoptosis were found and the result of flow cytometry showed that Interferon a and Tumor Necrosis Factor a could increase considerably the apoptosis rate(P0.01)and synthesis of DNA of tumor cells were inhibited obviously,the cell numbers in the S and G 2M phase were decreased,whereas the cells in the G 0/G 1 phase were increased, so the proliferation index(PI) were reduced.Conclusion Interferon aand Tumor Necrosis Factor a combined with chemotherapeuticdrugs could increase antitumor effect.
