The effect of angiotensin-(1-7) on expression of CD40/CD40L in THP-1 cells

Abstract

Objective To investigate the effects of angiotensin(1-7)[Ang-(1-7)] on expression of CD40,CD40L in THP-1 cells,which is stimulated by angiotensinⅡ(AngⅡ), identifying the protection of Ang-(1-7) in cardiopathy.Methods THP-1 cells were induced by fixed concentration of AngⅡand different concentration of Ang-(1-7), the CD40,CD40L expression on the macrophage were detected by flow cytometric analysis and CD40,CD40L mRNA were detected by RT-PCR.Results Ang-(1-7) can not only down-regulated the expression of CD40 and CD40L, but also the mRNA levels of CD40 and CD40L in cultured macrophage, in addition the Ang-(1-7) inhibitied them concentration dependently, the expression of CD40、CD40L step up again after using A-779 which is a specified blokiag agent of Ang-(1-7),and it is similar to the group of AngⅡ. Conclusion Ang-(1-7) inhibited the expression of CD40,CD40L dependently in THP-1 cells,which is stimulated by AngⅡ,the antagonism of Ang-(1-7) on the expression of CD40,CD40L is related to its specified acceptor-MAS.
