Study on Callus and Seedlings of Cichorium intybus

Abstract

In order to preserve the wild resources and meet the cultivated needs of people,taking the tender stems of Cichorium intybus as material,research was conducted by callus tissue culture,inducement differentiation,rooting of adventitious buds,rooting multiplication of tube seedlings,transplantation of tube seedlings and stable planting.The results showed that MS+ sucrose 30 g/L+BA 0.4 mg/L+2,4-D1.8 mg/L was the ideal medium for callus inducement and callus multiplication.1/2 MS +AgNO3 1.0 mg/L+ ZT 0.4mg/L+NAA 0.1 mg/L was the optimum medium for callus differentiation cultivation and adventitious buds cultivation.White+ABT 2 1.4 mg/L+sucrose 12 g/L+IAA 0.3 mg/L was the ideal medium for rooting of adventitious buds and rooting multiplication of tube seedlings.Transplantation of tube seedlings and stable planting were easy,and the tube seedlings stable planted maintained all botany characteristics of the wild Cichorium intybus.
