Abstract

Objective To use a high performance liquid chromatographic method for the direct resolution of sibutramine enantiomers with a α_1-AGP chiral stationary phase.Methods Influences of the pH values of the mobile phases,the organic modifiers,the buffer concentration,the flow rates and the column temperature on the enantiomeric separation were investigated.The enantiomeric separation was optimized.Results The best separation of enantiomers of sibutramine was obtained with isopropyl alcohol-20 mmol/L ammonium acetate buffer,pH 5.0(6∶94,V/V) as mobile phase with flow rate 0.6 mL·min~(-1) at 20 ℃.The detection wavelength was set at 223 nm.Conclusion Sibutramine enantiomers can be separated completely on α_1-AGP chiral stationary phase.The method is simple,rapid and reproducible.
