Abstract

Objective To know the temporal change of the activity of superoxide dismutase(SOD) and the concentration of malondialdehyde(MDA) in rat Sertoli's cells in cultured after exposed to PCB153.Methods Sertoli's cells were isolated from 18-to 20-day-old healthy male Sprague-Dawley rats,then were exposed to PCB153 at 0(solvent control,0.5% DMSO),10,20,30,40,50 μmol/L for 24,48 h,and a cell-free control group was established for the cell viability test,by using cell counting kit-8(CCK-8),and the cells were exposed to PCB153 at 0(solvent control,0.3% DMSO),10,20,30 μmol/L for 6,12,24,36,48 h for the determination of the activities of the SOD and the concentrations of the MDA.Results According to the results of CCK-8,10,20,30 μmol/L PCB153 were chosen to be the appropriate dosages to mensurate the activities of the SOD and the concentrations of the MDA.After 24 h of exposure,PCB153 at 20,30 μmol/L significantly decreased the activities of the SOD and increased the concentrations of the MDA compared with the control(P0.05),with a dose dependent manner.Compared with the previous time point of the same dose,the activities of the SOD were significantly increased after 12 h exposure to 30 μmol/L and 24 h exposure to 20 μmol/L(P0.05),and were significantly decreased after 36 h exposure to 10,20 μmol/L(P0.05).Meanwhile,the concentrations of the MDA were significantly increased after 24 h exposure to 30 μmol/L and 36 h exposure to 20 μmol/L(P0.05),and were significantly decreased after 48 h exposure to 20 μmol/L P0.05).Both of them had a time dependent manner at time points of 12,24,36 h.Conclusion In cultured rat Sertoli's cells,after being exposed to PCB153,the activities of the SOD may increase at the beginning and then decrease as time goes on,and decrease with the dose increase.the concentrations of the MDA may increase as time goes on,also increase with the dose increase.Both of them presents a dose-and time-dependent manner at 24 h.
