Abstract
Objective To explore the possible mechanism of manganese-induced apoptosis in PC12 cells.Methods Using PC12 cells as the model of dopaminergic neuron,MTT colorimetry test was used to detect the growth and survival state of PC12 cells in various doses of manganese chloride;apoptosis was detected by flow cytometry(FCM)and agarose gel electrophoresis(AGE);and immunocytochemistry was used to measure the expression and location of HSP70,Survivin,Bcl-2 and Bax.Results The results shown that manganese might suppress the proliferation of PC12 cells in dose and time-dependent manner,and might dose-dependently induce PC12 cells apoptosis;manganese chloride might also promote the expression of Bax in dose-dependent manners,which had a positive correlation to PC12 cells apoptosis(R1=0.972,P0.01);additionally,manganese chloride might dose-dependently inhibited the expression of HSP70,Survivin and Bcl-2 in PC12 cells as well,which had a negative correlation to PC12 cell apoptosis(R2=-0.990,R3=-0.976,R4=-0.980,P0.01).Conclusions Manganese can induce PC12 cells apoptosis,the enhanced expressions of Bax and the prohibited expressions of HSP70,Survivin,Bcl-2 may be the concrete pathway involved the mechanism of manganese-induced apoptosis of PC12 cells.
