Quality analysis of Radix Angelicae Dahuricae by HPLC

Objective:To develop a method for the determination of imperatorin and isoimperatorin for analysing the contents of the two coumarins in Radix Angelicae Dahuricae under different sampling area,different harvest time,different medicinal parts and different processed method.Methods:An HPLC method was adopted to determine the contents of two coumarins in Radix Angelicae Dahuricae.Chromatographic column:Shiseido C18 column(150 mm×4.6 mm,5 μm)was adopted;The mobile phase was methanol-water(60∶40),and the flow rate was 1.0 mL·min-1;The detecting wavelength was 300 nm and column temperature was 25 ℃.Results:Harvest time,medicinal parts and processed method were the major factors that influenced the contents of two coumarins.Conclusion:The method can be used to provide the experimental foundation for harvest time and the dryness of Radix Angelicae Dahuricae,and to assess the quality of Radix Angelicae Dahuricae.
