Abstract

Random amplified polymorphic DNA (RAPD) is a popular technique of fingerprinting. Good quality DNAs were extracted from 10 varieties accessions of tobacco by using improved “ROSE” method. Reproducible amplification products were obtained with rigorously optimized reaction conditions. Fifty\|three amplification fragments were generated by RAPD with 10 primers, among which 10 fragments were shared by all 10 accessions. The degree of polymorphism was 81.1%. RAPD patterns of 10 accessions generated by P 7 could be used as DNA fingerprint for identification of 10 tobacco varieties.
