Effects of up-regulated expression of microRNA-143 on cell proliferation and apoptosis of CasKi cells

Abstract

Purpose To investigate the effects of up-regulated expression of microRNA-143(miR-143) on cell proliferation and apoptosis of cervical squamous carcinoma cell lines CasKi.Methods Synthetic miR-143 mimics was transfected into CasKi cell line by lipofectamine.The expression of miR-143 in cervical squamous cell carcinoma tissues and the expression change of miR-143 in the CasKi cell line was measured by real time RT-PCR.Cellular growth and proliferation capacity were assayed by the methyl thiazolyl tetrazolium(MTT) assay and flow cytometry(FCM).The apoptotic rate was tested by TUNEL assay.Results The expression of miR-143 in CasKi cell and cervical squamous cell carcinoma tissues was down-regulated,in contrast to normal cervical tissue,and two carcinoma samples were non expression of miR-143.Compared with that of blank control group,miR-143 expression in the miR-143 mimics group cell increased significantly;the cell growth inhibition of miR-143 mimics group was demonstrated for 48 hours after cells treated with transfection by MTT results;FCM analysis showed that cells at G0/G1 phase in the miR-143 mimics group were significantly increased,while cells at S phase were decreased(P0.05);the apoptotic rate(9.45±2.31)% in the miR-143 mimics group was higher than the negative control group(4.03±1.35)% and the blank control group(3.85±1.61)%(P0.05).Conclusions Up-regulated expression of miR-143 can not only inhibit the proliferation but also induce the apoptosis capacity of CasKi cell.
