Abstract
In this paper comparison of the four methods for isolating genomic DNA from cotton was studied. The results showed that both the modified SDS(sodium dodecyl sulphate) and CTAB(cetyl triethyl ammonium bromide) methods for isolating genomic DNA from cotton were better than the original SDS and CTAB methods. The Yield of genomic DNA was not only related to extraction methods, but also to the positions sampled from cotton plants. DNA yield of modified CTAB method was higher than that of modified SDS method. The yields of isolation of genomic DNA from the positions of cotton plants was in order: anther petalleave fiber. The cotton genomic DNA isolated by the modified methods could not only be digested by enzyme, but also to meet a demand for study of subsequent molecular such as SSR(simple sequence repeats) and AFLP(amplified fragment length polymorphism) analysis.
