Isolation and identification of the differentiation potential of rat bone marrow-derived mesenchymal stem cells

Abstract

In this study,bone marrow-derived mesenchymal stem cells( BM-MSCs) were isolated from Sprague-Dawley( SD) rat by using the whole bone marrow adherence method. To identify their osteogenic differentiation potential,BM-MSCs were cultured in osteogenic medium. Alkaline phosphatase staining was performed on the 7th day and there are alkaline phosphatase positive cells,and Alizarin Red S( ARS) staining was performed on the 21 st day and bone nodules formation showed up. To identify their adipogenic differentiation potential,BM-MSCs were cultured in adipogenic induction medium,then switched in adipogenic maintenance medium,and on the 10 th day oil red O staining showed lipid drop formation. It indicates that rat BMMSCs with multiple differentiation potential have been successfully isolated from SD rat bone marrow,which lays a foundation for the clinic and research using rat BM-MSCs as seed cells.
