Abstract
Objective To identify novel genes involved in spermatogenesis.Methods Testes cDNA samples from Balb/C mice of different postnatal days were performed with mouse whole genome Affymetrix Genechip to screen novel testis-specific genes.The tissues distribution of the testis-specific genes in mice organs was identified by RT-PCR.The characteristics of selected genes were analyzed by various bioinformatic tools.Results A novel testis-specific gene(GenBank Accession No:XM-156106) with a differential expression in the developmental stages of testis was identified.The full length cDNA of 1364 bp contained an open reading frame of 1 146 bp which encoded a putative protein of 381 amino acids,we named this gene TSC43.RT-PCR analysis revealed that TSC43 mRNA was exclusively expressed in mice and human testis.Cellular mapping prediction of TSC43 indicated that its protein would be expressed in nuclear and one putative domain was anchoring domain of cAMP-dependent type Ⅱ PK predicted at the protein site 332-377.Conclusion The expression of TSC43 corresponds to the appearance of spermatid,indicating TSC43 may be involved in testis spermatogenesis.
