Abstract
Objective:To observe the protective effects of ischemic preconditioning and P-selectin expression in renal ischemia/ reperfusion injury model.Methods: Rat model of ischemia was made by left renal artery occlusion. The rats were divided into three groups randomly: sham-operated group(group S),ischemia-reperfusion group(group IR),ischemic reconditioning group(group IPC).The left renal underwent 60 minutes ischemia and 24 hours reperfusion. The levels of Scr,BUN,Superoxide Dismutase (SOD) and Malondialdehyde(MDA) in each group were examined. In addition,pathological changes were also examined by light microscope. P- selectin expressions in kidney were tested by immunohistochemistry. Results:① Scr,BUN and MDA:The values in group IPC were significantly lower than that in group IR( P 0.05). ②Blood SOD: The values in group IPC were significantly higher than those in group IR( P 0.05 ). ③Morphology: In group IR patch necrosis of renal tubules and the ultrastructural damages were irreversible.Whereas,in group IPC scattered necrosis of renal tubules could be seen and the most of ultrastructural damages were reversible. ④ P - selectin: The expression of P - selectin in group IPC was lower than that in group IR.Conclusion:Ischemic preconditioning could protect kidney from ischemia/ reperfusion injury via scavenging oxygen free radicals,improving the SOD's activity and reducing the levels of MDA. The mechanism might be that the herbs inhibits the expression of P - selectin in the kidneys.
