Abstract

Objective To investigate the effects of Cdx2 overexpression on immune-related genes in gastric cancer cell line MGC803 by cDNA microarray.Methods The total RNA was extracted and purified from Cdx2 transfected MGC803 cells and control MGC803 cells.The cDNA obtained by reverse transcription polymerase chain reaction (RT-PCR) was labeled with Cy5 and Cy3 fluorescence as probes and were hybridized with gene chip containing 21522 human 22K gene expression profiles.Subsequently,the two signal images were scanned by LuxScan 10K/A dual pathways laser scanner and analyzed by LuxScan3.0 image analysis software.Results In the 21522 target genes,551 genes were screened out for different expression level between these two groups.Among them,302 genes were up-regulated and 249 were down-regulated.Furthermore,there were 7 immune-related genes showed differential expression,i.e,HLA-E,RRAS,ULBP2,HLA-G,HLA-C,CTSB and CTSL.Conclusion Cdx2 overexpression upregulates immune-related genes in gastric cancer cell line MGC803,which may be associated with biological behaviors of gastric cancer cell line MGC803.
