Determination of 3 Phenolic Acids in Xiaoshuan Tongluo Tablets by HPLC with Electrochemical Detection

Abstract

To establish a high performance liquid chromatography with electrochemical detection(HPLC -ECD) method for determination of protoeatechuie acid,caffeie acid and ferulic acid in Xiaoshuan Tongluo tablets. Method:HPLC-ECD was used.The separation was performed on Zorbax SB-C_(-18) (150mm×4.6 mm,5.0μm) column by gradient elution.The mobile phase consisted of CH_3OH-4% HAc.The flow rate was 1.0 mL·min~(-1). The detection was done at 0.7V and column temperature is 30℃C.Results:The linear ranges are 0.7-7.9μg· mL~(-1) (r=0.9991) for protoeatechuic acid,0.2-5.4μg·mL~(-1) (r=0.9996) for eaffeic acid and 0.02-24.0μg ·mL~(-1)(r=0.9994) for ferulic acid respectively.The average recoveries (n=6) were 99.1% (RSD=1.8%) for protoeatechuie acid,98.7% (RSD=2.6%)for caffeic acid and 98.4% (RSD=1.9%)for ferulic acid respective- ly.Conclusion:A rapid,sensitive and accurate method for the determination of protoeatechuic acid,caffeic acid and ferulic acid was found.The study may provide a scientific basis for the quality control of Xiaoshuan Tongluo tab- lets.
