Abstract

Pyruvate kinase gene is a key factor in glycolytic pathway,playing an important role in fatty acid synthesis of oli bearing crops.Full-length cDNA clone of pyruvate kinase gene was for the first time obtained by using methods of 5′ RACE and overlap extension PCR based on our constructed EST library of Camellia oleifera.It was 2114bp in length and contained a 1737bp ORF(open reading frame) encoding of 579amino acids,proteinic molecular weight is 63766.5Da,molecular formula is C2774H4470N784O875S30,which showed high similarity with those of other plant species.Pyruvate kinase phylogenetic tree analyses revealed that the Camellia oleifera PK had closer relationship with PK's from Ricinus communis,Populus trichocarpa and Vitis vinifera.
