Determination of Two Nucleoside Active Components in Four Species of Cordyceps by Capillary Zone Electrophoresis

Abstract

Cordycepin and adenosine are the main nucleoside active components in medicinal fungus belonging to Cordyceps(Fr.)Link.In order to find an easy and low cost method to determine the contents of these two components in different species of Cordyceps,a capillary zone electrophoresis (CZE)method for separation and measurement of cordycepin and adenosine was developed.This is the first report on the presence of cordycepin in Cordyceps kyushuensis Kobayasi.Under the buffer condition of 0.025mol/L borate (pH9.4),applied voltage of 20kV and the detector wavelength of 258 nm,baseline separation of the active components in four species of Cordyceps was achieved and they were quantitatively analyzed.The linear calibration equations of the two components are Y=79.257X-0.006 for cordycepin and Y=81.623+0.004 for adenosine respectively.In addition,the effects of extracting time on the contents of cordycepin and adenosine were also investigated.
