Abstract
Objective To ohsen'e the efficiency and side effects of a combination of clozapine and paroxetine in the treatment of negative symptoms in schizophrenia. Methods 66 patients were randomly divided into the group of clozapine combined with paroxetine and clozapine combined with placebo. The efficiency and side effects were assessed with the postive and negative symptom scale (PANSS)and treatment emergent symptom seale (TESS) before and 4,8and 12 weekends after the treatment. The serum concentrations of clozapine and norelozapine was measured with HPLC meantime. Results Total and negative scores of PANSS significantly decreased after treatment in the group of clozapine combined with paroxetine. And negative sares were significantly lower than that of the group of clozapine combined with placebo from 8 week after the treatment . Serum concentrations of clozapine and norelozapine were significantly higher in clozapine combined with paroxetine. Total scores of TESS was significantly lower in study guoup than that in control group at the time of 4,8 and 12 week endsafter the treatment. Conclusions A combination of clozapine and paroxetine can remarkably improve negative symptoms in schizophrenia and produce feweu side effects.
