Expression of TLR in leucocyte from patients with type 2 diabetes

Abstract

Objective To investigate the distribution of TLR and the levels of IL-6 and TNF-α mediated by TLR in leukocyte from patients with type 2 diabetes.Methods The expression of TLR in leukocyte from peripheral blood were determined by FACS in 30 cases of control group and 35 cases of patients with type 2 diabetes.The levels of IL-6 and TNF-α from isolated PBLC were studied before and after cultivation and activation with LPS and HKLM.Results (1) In 30 cases of contral group,TLR2 level in lymphocyte was(1.75±0.50)%,TLR2 level in monocyte was(28.38±13.92)%,TLR4 level in lymphocyte was(9.07±1.60)% and TLR4 level in monocyte was(58.97±6.39)%.In 35 cases of patients with type 2 diabetes,TLR2 level in lymphocyte was(2.12±0.84)%,TLR2 level in monocyte was(35.09±12.69)%,TLR4 level in lymphocyte was(10.10±1.64)%,TLR4 level in monocyte was(62.85±6.63)%.A statistic difference was observed between two groups(P0.01).Furthermore,TLR2 level was(2.79±1.08)%in granulocyte in control group,there was no difference compared with TLR2((3.06±0.72)%) in patients with type 2 diabetes(P0.05).TLR4 level(12.64±2.05)% in patients with type 2 diabetes was more than that in control group in granulocyte.(2) Following and before cultivation and activation,the concentration of TNF-α was significantly higher in patients with type 2 diabetes than that in control group(P0.01).Before activation,it was found that no marked difference existed in the concentration of IL-6 between in patients with type 2 diabetes and in normal group(P0.05),but a diverse result existed on following activation.The rising of the concentration of IL-6 and TNF-α before and after activation was significantly higher in patients with type 2 diabetes than that in normal group(P0.05)by using matching t test.(3) The levels of TLR2,TLR4 and IL-6,TNF-α following activation by ligands were positively correlated with the level of HbA1C and FMN(P0.05),but not FBG.Conclusion There are changes of TLR and cytokines mediated by TLR associated with innate immunity in patients with type 2 diabetes,and it is possible for the changes to induce inflammation.
