Abstract
Objective To investigate the TLR2-mediated immune function in peripheral blood mononuclear cells of patients with type 2 diabetes.Methods The expression of TLR2 protein in lymphocytes,monocytes and granulocytes in peripheral blood were detected by flow cytometry among 35 type 2 diabetes patients and 30 controls.The single nucleus cell was activated by HKLM after being isolated in vitro and cultured.The expression of TLR2 and the secretion of TNF-α was also tested.Results The expression of TLR2 in lymphocytes and monocytes were(1.75±0.50)% and(28.38±13.92)% in controls,lower than those in the diabetes group((2.12±0.84)% and(35.09±12.69)%),respectively(P0.05).After being activated by HKLM,the expression of TLR2 in both controls and diabetes groups were increased.The TLR2 expression in controls were significantly higher than that in diabetes group after being activated by HKLM(P0.05).The expression of TNF-α after being activated with HKLM were(83.67±40.87)pg/mL in controls and(40.82±32.52)pg/mL in diabetes group,markedly higher than those activated with PBS((14.32±9.87) pg/mL in controls and(18.36±11.27) pg/mL in diabetes group)(P0.05).The secretion of TNF-α in controls significantly increased comparing to that in diabetes group.Conclusion The TLR2 expression in nuclear cells is increased in diabetes patients,but the TLR2-mediated immune function is damaged.
