Abstract

Objective To locate the deletion areas probably harboring tumor suppressor genes (TSGs) in glioblastoma(GBM).Methods Twenty loci on chromosome 10 were examined to detect LOH in 21 cases of glioblastoma (GBM).PCR based microsatellite polymorphism analyses were performed to detect loss of heterozygosity (LOH)on chromosome 10,fluorescence-labeled primers and Perkin Elmer 377 DNA Sequencer were applied.Results Among all informative cases of GBM,85.7% displayed LOH on chromosome 10,81.0% of displayed LOH on 10q and 66.7% on 10p.57.7% of informative loci showed LOH in our series,in which the higher frequent LOH(60%) were observed in the following chromosomal region or loci:from D10s185 to D10s192 on 10q22.3-23.3,D10s519 to D10s249 on 10p14-15.1,D10s1693 to D10s212 on 10q24.2- 26.3 ,D10s547 on 10p12.2-14,D10s537 on 10q21.3.Conclusions Loss of genetic material on chromosome 10 may play an important role on the molecular pathogenesis of GBM.The chromosomal regions from loci D10s185 to D10s192 on 10q 22.3 -23.3,D10s591 to D10s249 on 10p14-15.1,D10s1693 to D10s212 on 10q24.2-26.3,and at loci D10s547 on 10p12.2-14 and D10s537 on 10q21.3 may harbor novel tumor suppressor genes associated with GBM.
