Abstract
OBJECTIVE To observe the inhibitory effect of Combretaftation A4 prodrug( CA4P ) on human cervical carcinoma Hela cell in vitro and in nude mice. METHODS The inhibitory effect of CA4P human cervical carcinoma Hela cell was investigated by MTT analysis. Hela cells were inoculated subcutaneously into nude mice. Volumes of tumor were measured and growth of tumor was observed and the inhibitory rate of tumor growth was computed. RESULTS After treated with CA4P for 24 or 48 hours,MTT assay showed the dose range from 0. 5μg·mL -1 to 64 μg·mL -1 had the inhibitory ratio range from 15% to 75% and from 18% to 80% respectively. The IC50 was 0. 336 μg·mL -1 and 0. 207 μg·mL -1,respectively. The inhibitory rate of tumor growth in nude mice carried tumour was 30% to 50% ,and it was related with the dose. CONCLUSION CA4P could inhibit the growth of human cervical carcinoma Hela cell in vitro and in vivo.
