Abstract

A cellulase-producing strain was isolated by the congo red dying method on the culture plates which has sodium carboxymethyl cellulase.The stain was identified as Actinomycete.The maximum activities of carboxymethyl cellulase enzyme of CMC-Z were increased via series single factor and orthogonal comparisons.The optimal culture medium and conditions for enzyme production by CMC-Z were obtained.The optimum of carbon source and nitrogen source were soluble starch 1% and gelatin 2%,pH of culture medium 7.0;inoculum rate 7.5%,culture temperature at 30 ℃,fermentation time for 72 h.The results of range analysis and variance analysis showed that the order were the soluble starchpHloading capacitygelatineinoculum sizetemperaturefermentation time.The maximum activities of CMC was 38.37 u·mL-1 under the optimized fermentation conditions.
