Abstract
Objective:To compare the differences of type 1 diabetes mellitus(T1DM)SD rat models induced by different doses of streptozotocin(STZ)and to optimize the dose for establishment of T1DM models. Methods:Eighty male SD rats were randomly divided into 4 groups including 3 diabetic groups which were injected with 40,50,and 60 mg/kg of STZ(groups D40,D50,and D60)once through caudal vein respectively, and 1 control group which was injected with citrate buffer solution of the same volume. The fasting blood glucose, urine glucose, and daily water intake, urinary output, and food intake, and body weight of rats in all the groups were observed. Results:The model success rates of groups D40,D50,and D60 were 70.0%,100.0%,and 100.0%,and the death rates were 0.0%,10.0%,and 60.0% respectively. All of the rats in the 3 groups showed clinical manifestation of diabetes mellitus. The fasting blood glucose and urine glucose levels had downtrend in group D40 in 2 weeks, but kept high level in groups D50 and D60.Conclusion:The dose of 50 mg/kg of STZ is the best amount for establishment of T1DM rat models by injection through caudal vein which leads to higher success rate, lower death rate, and stable clinical features.
