RP-HPLC simultaneous determination of chlorogenic acid,baicalin, phillyrin,baicalein and wogonin in Dawei granules

Abstract

Objective:To establish an RP-HPLC method for determination of chlorogenic acid,baicalin,phillyrin,baicalein and wogonin in Dawei granules.Methods:The HPLC separation was achieved on a Diamonsil C18(200 mm×4.6 mm,5 μm)column at 35 ℃.The mobile phase consisted of acetonitrile-0.05% phosphoric acid solution with gradient elution(0-10 min,10%A→15%A;10-15 min,15%A→25%A;15-25 min,25%A→28%A;25-35 min,28%A→35%A)at a flow rate of 1.0 mL·min-1.The UV detection wavelength was set at 230 nm.Results:The linear ranges were 2.80-28.0 μg·mL-1(r=0.9992),25.71-257.1 μg·mL-1(r=0.9994),0.71-7.1 μg·mL-1(r=0.9995),0.81-8.1 μg·mL-1(r=0.9990),0.50-5.0 μg·mL-1(r=0.9992),respectively.The average recoveris(n=9)were 100.0%,99.0%,99.0%,99.9% and 99.4%,respectively.Conclusion: The method is simple,accurate and repeatable,and can be used for quality control of Dawei granules.
