Abstract
Objective To explore the effect of transforming growth factor alpha (TGF α) on cell cycle of gastric carcinoma cells, and investigate the role of tyrphostin 51 in inhibiting TGFα activity Methods Human gastric adenocarcinoma cell line SGC7901 were cultured in vitro and treated with TGFα. to observe the cell proliferation. The cells were counted daily for 8d. Synchronization of cell cycle at G1-phase was Achieved before the administration of TGFα treatment, and the analysis of the changes in cell cycle using flow cytometry (FCM) and the assay of the intracellular DNA synthesis by ~3H-TdR incorporation were performed The changes in cellular apoptosis was observed using FCM after the cells were treated with tyrphostin 51 Results TGF α accelerated the cell proliferation and upregulated the growth of SGC7901. The S-phase cell percentage and the G2+M-phase cell percentage were obviously increased after the treatment with TGFα. resulting also in the obvious enhancement of DNA synthesis. The cellular apoptosis was obvious after the cells were treated with tyrphostin 51 for 48 h, Conclusions TGF αcan promote cell proliferation and obviously accelerate the cell cycle of human gastric adenocarcinoma cell line SGC7901.Tyrphostin 51 can induce cellular apoptosis by inhibiting the PTK activity of TGFα recepter
