Abstract
Objective To study the inhibitory effect of pueraria isoflavones on prostate hyperplasia induced by testosterone in emasculated rats and clarify its mechanism. Methods 72 male Wistar rats were randomly divided into 6 groups,the control group( only Normal Saline,NS),the finasteride group[1. 0 mg /( kg·d) ],the model group,the pueraria isoflavones group[180,120,60mg /( kg·d) respectively]. The rats in the control group underwent sham operation,the rest were emasculated operation. From the 8th day,except the blank control group,rats in the above-mentioned 5 groups were given Testosterone Propionate [10 mg /( kg·d) ] by subcutaneous injection for continuous 10 days to copy prostatic hyperplasia model. Then maintaining with subcutaneous injection once every two days,the blank control group and model group were given normal saline[1. 0 mg /( kg·d) ],the finasteride group was given finasteride [1. 0 mg /( kg·d) ],the pueraria isoflavones groups were given for 35 days. On the 36th day,serum was taken and the PAP vitality was determined. Then the prostate was separated,and weighed,the index was calculated,prostate volume change using the toe volumetric measuring instrument and the histology and morphology of the prostate was observed. Results Pueraria isoflavone in different dose groups had significant effect on wet weight,volume and index of rat prostate( P 0. 05). And alleviated the change of prostate caused by Testosterone Propionate: epithelium thinning,luminal secretion and interstitial substance decreased. Conclusion Pueraria isoflavone had inhibitory effect on benign prostatic hyperplasia in rats.
