Experimental study on the method of nourishing Yin and dispersing stagnant liver qi in the treatment of liver fibrosis in rats

Abstract

Objective To investigate the therapeutic effect of method of nourishing yin and dispersing stagnant liver qi on hepatic fibrosis in rats. Methods 28 SD male rats were randomized into the normal group,model group,positive drug group and Chinese herbal medicine group of 7 in each group. Except the normal group,carbon tetrachloride( CCl4)-induced model of hepatic fibrosis was established in other three groups. After modeling,the positive drug group was given colchicines of 0. 25 mg / kg with gavage method,the Chinese herbal medicine group was given Yiguanjian concentrated liquid of 1 m L /100 g with gavage method,while the normal group and the model group were given equal amount of physiological saline once a day. The body weight of rats was monitored every week. After 4 weeks,the weight of body,liver and spleen,and the contents of hydroxyproline in liver tissue of rats were detected,the liver weight index and spleen weight index calculated,and the condition of liver fibrosis evaluated by pathological staining. Results Compared with the normal group,the condition of hepatitis and hepatic fibrosis became aggravated( P 0. 01),the body weight decreased,the liver index and spleen index raised( P 0. 01),liver hydroxyproline increased( P 0. 05,P 0. 01) in the model group. Compared with the model group,the condition of hepatitis and hepatic fibrosis became alleviated,the spleen weight index and liver hydroxyproline were decreased( P 0. 01) in the positive drug group and Chinese herbal medicine group; the liver weight index dropped and the body weight increased,but the difference had no statistical significance( P 0. 05). Conclusion The method of nourishing Yin and dispersing stagnant liver qi can reduce the degree of hepatic fibrosis in rats induced by carbon tetrachloride and alleviate the liver and spleen enlargement.
