Effects of "Xiezhuo Jiedu Decoction" on expressions of TLR4,IκB-α and IL-8 in ulcerative colitis rats

Objective According to the pathogenesis of ulcerative colitis(UC),we composed Xiezhuo Jiedu Decoction,which acts to purge turbidity and ascend lucidity,detoxify and remove blood-stasis.We carried out the animal experiment to observe the effects of Xiezhuo Jiedu Decoction on TLR4,IκB-α and IL-8 in UC rats,in an attempt to study its possible mechanisms.Methods Forty Wistar rats were randomly divided into 5 groups with 8 rats in each group: blank group(BG),model group(MG),SASP group(SG),low-dose Xiezhuo Jiedu Decoction group(LG) and high-dose Xiezhuo Jiedu Decoction group(HG).Except BG,the other 4 groups were given 2,4,6-trinitrobenzene-sulfonic acid and TNBS/ethanol to duplicate ulcerative colitis in rats.On the fourth day after model establishment,BG and MG were given 0.9% NaCl 10ml/kg by gavage;SG,LG and HG were respectively given SASP at 0.3g/kg,Xiezhuo Jiedu Decoction at 10.0g/kg and 20.0g/kg by gavage,once a day for 2 weeks.The levels of TLR4 and IκB-α were detected by immunohistochemical staining and the content of serum IL-8 by radioimmunoassay.Results Compared with BG,the levels of TLR4 and IκB-α and the content of serum IL-8 were significantly increased in MG(P0.01);compared with MG,the levels of TLR4 and IκB-α and the content of serum IL-8 were strikingly decreased in SG,LG and HG(P0.05,P0.01);compared with SG and LG,the levels of TLR4 and IκB-α and the content of serum IL-8 were obviously lower in HG(P0.05,P0.01).Conclusion Xiezhuo Jiedu Decoction can reduce the levels of TLR4 and IκB-α and the content of serum IL-8 to fight against ulcerative colitis.
