Abstract
The determination of chestnut polysaccharide content using anthrone-sulfuric acid method was studied and the operating conditions were optimized.The results showed that the optimal operating conditions were:4 mL of 0.2% anthrone-sulfuric acid solution,heated in boiling water bath for 3 min,cooled with city water for 40 min,read the absorbance under 600 nm wavelength in 10 min after cooling.The anthrone-sulfuric acid solution must be used quickly.Polysaccharide concentration between 0.02~0.06 g/L was the best for determination.The RSD in accuracy experiment was 0.792%,and the average recovery rate was 107.2% with RSD of 4.6%.The content of polysaccharide in chestnut determined by this method was 12.67%.
