Abstract
In this study, crude polysaccharides isolated from Chaenomeles sinensis was purified using 10% trichloroacetic acid solution. The content of polysaccharides was determined by phenol-sulfuric acid method and calculated from the conversion coefficient of purified polysaccharide to glucose. The optimal measure conditions were determined by investigating the relationship of UV absorbance of polysaccharides at 490 nm with action time and temperature of phenol-sulfuric acid system on polysaccharide and amount of phenol and sulfuric acid. The optimal conditions were obtained as follows:the action temperature 100℃,the action time 15 min,the amount of phenol 1.0 mL,the amount of sulfuric acid 5.0 mL.The average content of polysaccharides of 12.16% was obtained under the optimum testing conditions.
