Abstract
Objective to explore the action of main reagent alkaline lysis method of extracting plasmid DNA.Methods adopt different SolutionⅠ、SolutionⅡ、SolutionⅢ to extract DNA in the plasmid.Results the quantity of plasmid DNA is obviously decreasing by method three,the protein of the plasmid DNA is more than the other methods by method four.Conclusions solutionⅠhas little influence to the experiment result,and NaOH in the solutionⅡ relates to the quantity of extracting plasmid DNA, KAc in the SolutionⅢ is one of the important factors in the protein,s purification.
