Abstract
Objective To study the effect of trimetazidine or ramipril alone and their combination on myocardial cell apoptosis and Caspase-3expression in chronic heart failure rats.Methods Fifty male Wistar rats were randomly divided into sham operation group,model group,trimetazidine group,ramipril group and combined treatment(trimetazidine and ramipril)group(10in each group).A chronic heart failure model was established by suprarenal aortic constriction.LVEDP and±dp/dtmaxwere recorded,AI of myocardial cells was assayed by TUNEL,expressions of Caspase-3protein and mRNA were detected by SP immunohistochemistry and RT-PCR respectively in different groups.Results The LVEDP,AI and the expression levels of Caspase-3protein and mRNA were significantly lower in trimetazidine group,ramipril group and combined treatment group than in model group(0.14±0.01 vs 0.22±0.02,0.14±0.02 vs 0.54±0.02,0.08±0.02 vs 0.54±0.02,and 0.32±0.02,0.91±0.02,P0.01)while the±dp/dtmaxwas significantly higher in trimetazidine group,ramipril group and combined treatment group than in model group.Conclusion Trimetazidine or ramipril alone and their combination can effectively improve cardiac function and inhibit myocardial cell apoptosis in chronic heart failure rats.The therapeutic effect of combined trimetazidine and ramipril is better than that of trimetazidine or ramipril alone.
