Abstract
Objective To study the effect of trimetazidine and ramipril on expression of TGF-β1 in myocardial tissue of chronic heart failure(CHF)rats.Methods A CHF model of Wistar rats was estabablished by constricting the suprarenal abdominal aorta.Fifty Wistar rats were randomly divided into model group,trimetazidine treatment group,ramipril treatment group,combined treatment group and sham operation group(10in each group).The left ventricular mass indexes(LVMI),left ventricular end diastolic pressure(LVEDP)and maximum left ventricular pressure rising and falling(±dp/dtmax)were measured.Myocardial morpholy of left ventricle was observed under optical microscope with Masson staining.Expression of TGF-β1 and TGF-β1 mRNA in left ventricle was detected by SP immunohistochemistry and RT-PCR,respectively.Results The ±dp/dtmax was significantly higher,whereas the LVEDP and LVMI,the expression level of TGF-β1 and TGF-β1 mRNA were significantly lower in trimetazidine treatment group,ramipril treatment group and combined treatment group than in model group.Optical microscopy showed that the myocardial damage was less in trimetazidine treatment group,ramipril treatment group and combined treatment group than in model group.Conclusion Trimetazidine or ramipril alone and their combination can improve the heart function and myocardial fibrosis in CHF rats.The effect of combined trimetazidine and ramipriis is better than that of trimetazidine or ramipril alone.
