Application of PEG/(NH_4)_2SO_4 Aqueous Two-phase System to Extraction of Total Flavone of Populus Canadensis Moench.Leaves

Abstract

Objective:To find an effective method for extraction and purification of total fiavone of Populus Canadensis Moench. leaves.Methods:In the experiment,extraction and content determination of flavone of Populus Canadensis Moench.leaves were carried out simultaneously by using aqueous two-phase extraction partitioning and UV-spectrophotometry method.Results:This aqueous two-phase system was prepared by dissolving suitable amounts of polyethylene glycol (PEG)and (NH_4)_2SO_4 in aqueous solution.Flavone of Populus Canadensis Moench.leaves were recovered with optimum high yield and high concentration in the upper polymer-rich phase when 25% PEG 400 and 12%(NH_4)_2SO_4 were applied.The optimum extraction conditions were pH value 9,NaCl3 %,temperature 25℃and leaching solution volume 3 mL.The precision (RSD)was 0.28% (n=5).Conclusion:The method is proved to be good in repro- ducibility,simple and convenient in operation,and it is effective to extract flavone.
