Effect evaluation of chocolate agar media made of horse,rabbit and sheep blood using for isolate haemophilus influenzae

Abstract
Objective To compare the isolate rates of haemophilus influenzae in sputum by chocolate agar media made of horse,rabbit and sheep blood,and the growth of haemophilus influenzae on the three kinds of chocolate agar media.Methods Sputums and Haemophilus influenzae ATCC10211 were streaked on the three kinds of chocolate agar media and incubated at 36℃ in a candle jar for 24 hours.The isolate rates of haemophilus influenzae by three kinds of chocolate agar media and the growth of haemophilus influenzae ATCC10211 were compared.Results The isolate rates of Haemophilus influenzae in sputum by chocolate agar media made of horse,rabbit and sheep blood were 34.3%,35%,8.3% respectively.Haemophilus influenzae ATCC10211 grew very well on chocolate agar media made of horse and rabbit blood,its diameter of colony was 0.5～1.0mm.But it did not grow well on the chocolate agar media made of sheep blood,its colony was as small as a sand and hard to be seen with naked eye.Conclusion Chocolate agar media made of horse and rabbit blood are better than the chocolate agar media made of sheep blood in haemophilus influenzae isolation in sputum.
