Analysis of Factors Influential to Adventitious Bud Regeneration of Isolated Leaves of Plum ‘Damas1869’

Abstract

Experiment used the isolated young leaves of test-tube seedlings of plum ‘Damas1869’to study the effects of different cultivation conditions on adventitious bud regeneration,the results indicated that F14 medium was the optimal basic culture medium for the adventitious bud regeneration and it was superior to MS and WPM medium.The optimal hormone contents were TDZ 4 mg/L + IBA 0.5mg/L.The regeneration medium containing agar 4.5 g/L was a very effective method to improve adventitious bud regeneration from the leaves whose adaxial side touched to the medium.Cultivation under light after 5 days' cultivation under dark could enhance the regenerative rate,the regenerative rate was 73.5% or so and 3~5 buds per leaf could be regenerated on average.
